Aseptic Technique Basics

Practical contamination prevention habits for cell culture and routine bench work

Prepare the work area before opening materials. Reduce unnecessary movement, keep items organized, and avoid crossing clean materials over waste or used tips.

Handling sequence

Move from clean to less clean tasks. Avoid returning to sterile reagents after touching waste, incubator handles, notebooks, or non-sterile surfaces without changing gloves or re-cleaning.

Bottle and tube behavior

Keep lids open for the shortest practical time. Do not place caps on uncertain surfaces. Avoid touching inside surfaces, threads, or pipette tips to anything outside the intended sterile path.

Practical habits

  • Plan the whole sequence before starting.
  • Keep only necessary items in the hood.
  • Wipe bottles and racks before use when appropriate.
  • Watch where pipette tips travel.
  • Label clearly before opening cells or reagents.
